• Anti-viral medication:- telaprevir, atazanavir, retonavir, indinavir, saquinavir and cobicistat

• Recreational drugs:- amyl nitrate (poppers)

Drug Interaction of Tadalafil Tablets

Drug-Drug Interaction

Chloramphenicol

The use of this drug is contraindicated because the use of chloramphenicol will increase the effect of Sildenafil by affecting the patient hepatic and intestinal enzyme metabolism.

Isosorbide dinitrate

The use of this drug is contraindicated because the combination of this drug either increases the effects of the other by vasodilation which leads to fatal hypotension.

Nitroprusside sodium

The use of this drug is contraindicated because the combination of this drug either increases the effects of the others by pharmacodynamic synergism.

Enzalutamide

Use alternative drugs because the use of enzalutamide will decrease the effect of Sildenafil by affecting patient hepatic and intestinal enzyme metabolism.

Acetazolamide

Therapy should be administered with caution because the use of Sildenafil increases the effect of acetazolamide by pharmacodynamic synergism and it also increases the risk of hypotension.

Drug-Food Interaction

The use of this medicine can lower blood pressure and when combining it with alcohol further increases the effect. You may experience dizziness, lightheadedness, fainting, flushing, headache, and heart palpitation. Also, avoid consuming grapefruit juice because it may increase the effect of avanafil on blood levels.

Drug-Disease Interaction

Cardiovascular disease:

The use of phosphodiesterase-5 enzyme inhibitors is contraindicated in patients with cardiovascular disease. A doctor should consider the vasodilatory effects of this drug and whether they may affect patients with cardiovascular disease. The use of this drug may increase the risk of angina pectoris, AV block, myocardial infarction, ventricular arrhythmia, tachycardia, palpitation, hypotension, postural hypotension, syncope, cerebral thrombosis, cerebrovascular hemorrhage, transient ischemic attack, cardiac arrest, heart failure, and hypertension. These events can occur during or after sexual activity.

Renal dysfunction:

Therapy should be administered with caution in patients with severe renal disease or on renal dialysis. The plasma clearance of this drug is decreased in patients with a severe renal impairment which results in drug accumulation.

Liver diseases:

The use of this drug is not recommended for patients with severe hepatic impairment because the pharmacokinetic disposition of this drug has not been assessed in patients with severe hepatic impairment.

What are the potential drug-drug interactions of Tadalafil Tablets

The use of this medicine can impair the absorption, metabolism, and elimination of carbohydrates, fatty acids, lipids, and fatty-acylglycerol products. Therefore, it is recommended to take an antacid separated and broken down by at least three minutes before taking Tadalafil Tablets.

Increased risk of intracranial hypertension:

The increased cardiovascular risk of Tadalafil may occur in patients with a history of heart attack or stroke because the increased incidence may be influenced by these factors. The increased cardiovascular risk may occur only in the first few months of taking this medication and may occur following cessation of heart disease or before the start of treatment with other medications.

Therapy should be administered with caution in patients with renal impairment and in patients with severe renal disease because the patients with renal impairment may experience increased levels of blood in the urine. Therapy should be administered as needed to achieve the goal of decreased stole urine.

Drugs Adverse Reactions
Received the “Gold Standard” in 2020. For the next five years, Accord had “very little to say” about patient health but, in June 2017, the company received approval to carry out randomised controlled trials (RCTs) of tadalafil in patients with erectile dysfunction (ED). In June, Accord published in the peer-reviewed journalJAMA(“the first oral medication to treat erectile dysfunction – a potentially life-threatening condition – that has been shown to be safe and effective in clinical trials”). In October, Accord published in the journalLancet(“the first non-prescription medication to treat erectile dysfunction – a potentially life-threatening condition – that has been shown to be effective in clinical trials”). Accord conducted three RCTs in 50 Accord-naive patients (aged 18 to 64 years) to evaluate the safety and effectiveness of tadalafil in these 50 Accord-naive patients. A total of 21 Accord-naive patients (aged over 64 years) met the criterion for RCT. In these patients, tadalafil was applied three times daily (every 4 hours) for 5 days. Accord and its RCT partner, highstreet pharmaceuticals, conducted four investigational RCTs in 10 Accord-naive patients (aged over 18 to 64 years) to evaluate the efficacy and safety of tadalafil in these 10 Accord-naive patients. In these patients, tadalafil was applied three times daily for 5 days.
